Ubiquitin(Ub)isa76aminoacidproteinwidelyexpressedinthecytoplasmicandnucleusofcells.UbisposttranslationallyconjugatedtoproteinsbytheE1,E2,E3proteinubiquitinationcascade.UbcanbeconjugatedonproteinsasmonoUborpolyUbchains.Proteinubiquitinationplaysbothproteolyticandnonproteolyticfunctions.Usually,polyubiquitinatedproteinsaretargetedtothe26Sproteasomeforproteolysis.Typicalconcentrationtosupportinvitroubiquitinationis50-100µM.InUb(K11only),alllysineresiduesbutlysine11weresubstitutedwitharginines,therebyitonlysupportstheformationoflysine11-linkedpolyubiquitinchains.
